Cryo-EM reveals how aphidicolin blocks human DNA polymerases

A bioRxiv preprint reports cryo-EM structures showing aphidicolin blocking human DNA polymerases δ and ε at the site where they add DNA building blocks. Reconstituted assays and the structures suggest that differences in how the enzymes grip the DNA template help explain why Pol δ is more sensitive.

THIK1 Structures Show Related Inhibitors Act Differently

A bioRxiv preprint reports cryo-EM structures of human THIK1 bound to two chemically related inhibitors. Both occupy the same pore site, but structural and electrophysiological results indicate that they affect a second channel gate differently.

Split-GFP graphene grid captures tagged proteins for 2.58 Å cryo-EM

A bioRxiv preprint presents SGFP–G, a split-GFP graphene grid that selectively captures GFP11-tagged proteins and lets researchers check enrichment by fluorescence before vitrification. The authors demonstrated the platform with apoferritin and obtained a 2.58 Å cryo-EM reconstruction.
